The editors (both of the U. of Vermont's Department of Pathology) present an "eclectic" collection of what they consider state-of-the- art methods for imaging cells and molecules. The 21 papers begin with light microscopic methods for observing mRNA, calcium, and collagen. Next come contributions discussing confocal scanning laser microscopy, quantitative computer-assisted image analysis, laser scanning cytometry, laser capture microdissection, microarray image scanning, near-field scanning optical microscopy, atomic force microscopy, and reflection contrast microscopy. Three dimensional analysis of single particles by electron microscopy is discussed in a pair of chapters dealing first with sample preparation and data acquisition, and then with image processing. The volume concludes with a look at new microbiopsy system that enables rapid preparation of tissue for high-pressure freezing. The protocols follow the series format, offering step-by-step laboratory instructions, discussion of the principles behind the technique, lists of equipment and reagents, and advice on troubleshooting and pitfalls.
