This book belongs to the well-known series Methods in Molecular Biology, mainly devoted to procedures and methodologies. It collects together protocols for the separation, purification and sequencing of peptides and proteins. It is composed of thirty short chapters written by specialists in their fields. The first twelve chapters dealt with separation and purification methods of peptides, with or without preliminary enzymatic or chemical cleavages. They include electrophoresis (gel or capillary), electrobiotting and HPLC. Then come three other chapters on aminoacid analysis. Use of mass spectrometry is described in three chapters with details on the usable modes: fast-atom bombardment (FAB), electrospray (ES) or matrixassisted laser-desorption ionization (MALDI), culminating in the association of manual stepwise Edman degradation and MALDI-MS in the so called 'Protein ladder sequencing method', first published in 1993. There then follow four chapters on N-terminal peptide sequencing, including dansyl and Edman methods, and three chapters on C-terminal sequencing, either manually or automatically. Finally, more specialized situations including analysis and identification of sites of phosphorylation, glycosylation, etc are described. The book is full of valuable information, mainly at the practical level. Additional notes are clear and references are recent and judiciously chosen. I warmly recommend this book to all those who work in peptide and protein chemistry.
